This method specifies a procedure to extract and quantitate the Soybean Agglutinin (SBA) protein, also known as soybean lectin, by an Enzyme-Linked-Lectin Assay (ELLA). The assay utilizes the carbohydrate specificity of lectins by using polyacrylamide linked carbohydrates for capture and biotin labeled carbohydrates for detection. Specifically it uses a polyacrylamide-linked N-acetylgalactosamine (GalNAc-PAA) for capture and the biotinylated version (GalNAc-PAA-Biotin) for detection. To ensure optimal extraction of the protein and reliability of assay results, perform the procedures according to the parameters specified. Applicable to measuring the soybean anti-nutrient, lectin, in soybean grain.
