Test portions are extracted by heating with acetic acid solution. Extracts are cleaned up using solid-phase extraction (SPE) C18 cartridges. After periodate and peroxide oxidation, they are analyzed by highperformance liquid chromatography (HPLC) with fluorescence detection. Most toxins (STX, C1,2, B-1, dcSTX, and GTX2,3) can be quantified after simple SPE-C18 cleanup. Extracts containing the toxins NEO, GTX1,4, C3,4, and B-2 must be further purified by using SPE-COOH cleanup/separation. PSP toxin concentrations are calculated by comparison of peak areas or heights in the test samples with those of the standards.
